Poker Face: Flotsam is a Witcher 2 Assassins of Kings Enhanced Edition side-quest you can finish during the first chapter, when you visit Flotsam.
Those who manage to complete this specific quest, will earn additional Orens as well as other rewards.
Poker Face: Flotsam is a simple side-quest and shorter than other quests available in the game; however, make sure you finish it before the Troll Trouble side-quest.
To begin Poker Face: Flotsam visit the inns basement. Look for a character named Bendeck who is sitting at a dice poker table near a wall. Talk to him to activate Poker Face: Flotsam.
Quest Description
“When Geralt was staying in Vizima, he was less known among the gamblers as a witcher and more as a consummate dice poker player. His fame spread across all of Temeria and there were bound of people in Flotsam who would recognize him as a gaming partner.
Geralt was immediately invited for a game with two local players. He also learned about the third – the local champion, a man who wouldnt play with just anyone.”

In Witcher 2 Assassins of Kings Enhanced Edition the rules that must be followed to win a dice poker game are similar to those of a Yahtzee game mixed with those of a poker game. You have 5 dices which must be rolled during the first phase.
When the second phase starts, you will be asked to re-roll the dices you dont need. Notice the signs on the dices and try to get the most powerful combination, 5-of-a-kind.
You can also re-roll 2 or even 5 dices. Additionally, you can try to get 4-of-a-kind, two pairs, one pair, etc Make sure that the dices fall on the table, and be careful when you place your bets.
To finish the quest, first beat Bendeck, then, at the same table, beat Casimir. Your third opponent should be Zindrab, at the same table. If you manage to defeat him, you will be allowed to play against Einar Gausel in the non-human district. Defeat him and he will tell you that the best poker dice player is Sendler.
You will find Sendler in Lobinden. Play until you defeat him and you will complete the Poker Face: Flotsam side-quest.
After you beat Sendler, you can get the coins you have won, or you can ask him to give you a trap pattern.
Note: While inside Sendlers house, observe the troll head on the wall. The item plays an important role in the Troll Trouble side-quest.
